Mission 02 · MOTS-c / research guide
A small message.
A long way to go.
MOTS-c comes from the cell's energy machinery. The leap from an interesting signal to a useful treatment is still ahead.
About 3 min · Study settings kept in view

The short version.
Added MOTS-c improved physical performance in mice. Human studies in this collection mostly observe the body's own MOTS-c; that does not test a treatment.
- The setting
- Cells, mice & a human cohort
- The catch
- A marker isn't a treatment
- Missing
- Published human treatment results

The powerhouse sends a message
Mitochondria help cells make usable energy. They also carry the genetic instructions for MOTS-c, a peptide made of 16 amino acids, the building blocks of proteins.
In experiments with stressed human and mouse cells, MOTS-c moved into the nucleus, the cell's main store of genetic instructions, and helped regulate stress-response genes. Think of this as a message within a cell, rather than evidence that an injection helps a person cope with stress.

The moving mouse is a real finding
In the mouse performance study, added MOTS-c improved physical performance. Older mice improved on running, grip and gait measures. In a separate part of the same paper, exercise raised naturally produced MOTS-c in people.
Those are two different observations: exercise changing the body's own peptide, and researchers giving extra peptide to mice. Neither establishes an exercise replacement or better stamina in people.
A later CK2 study found a direct molecular target. CK2 is an enzyme; its activity rose in mouse muscle but fell in fat. Even within one body, “more activity” was not the whole story.

The human paper did not give anyone MOTS-c
A study of people receiving chronic hemodialysis measured the MOTS-c already circulating in their bodies. Higher levels were associated with a combined outcome of death and non-fatal cardiovascular events.
Nobody received MOTS-c as a treatment. A marker may accompany illness without causing it. This result cannot tell us what giving the peptide would do, or predict the same association in people without that kidney disease.

What is missing for people?
FDA's review of MOTS-c found no clinical studies establishing the effects of giving it to people, and no clinical data on how the body handles it. A registered MOTS-c trial had no posted results as of September 30, 2026. A registration is a plan, not an answer.
There is no established human benefit or safety result here for added MOTS-c. The review of stress and metabolism research is a useful map of the biology, but animal experiments cannot supply a personal treatment plan.
Next flight path NAD+: a blood result and the bigger questionThe body making a peptide and a person receiving extra peptide are different experiments.
